Abstract
La presente invención se refiere de manera general a inmunoconjugados, en particular inmunoconjugados que comprenden un polipéptido interleuquina-2 mutante y un anticuerpo que se une a PD-1. Además, la invención se refiere a moléculas de polinucleótido codificantes de los inmunoconjugados y a vectores y células huésped que comprenden dichas moléculas de polinucleótido. La invención se refiere además a métodos para producir los polipéptidos mutantes, a composiciones farmacéuticas que los comprenden y a usos de los mismos. Reivindicación 1: Inmunoconjugado que comprende: (i) un anticuerpo que se une a PD-1 e (ii) un polipéptido que señaliza mediante IL-2Rbg, en particular un polipéptido IL-2 o un polipéptido IL-15, en el que el polipéptido IL-2 es un polipéptido IL-2 mutante, en el que el polipéptido IL-2 mutante es una molécula de IL-2 humana que comprende las sustituciones de aminoácidos F42A, Y45A y L72G (numeración respecto a la secuencia de IL-2 humana SEQ ID Nº 19) y en el que el anticuerpo comprende (a) una región variable de cadena pesada (VH) que comprende una RHV-H1 que comprende la secuencia de aminoácidos SEQ ID Nº 8, una RHV-H2 que comprende la secuencia de aminoácidos SEQ ID Nº 9 y una RHV-H3 que comprende la secuencia de aminoácidos SEQ ID Nº 10 y (b) una región variable de cadena ligera (VL) que comprende una RHV-L1 que comprende la secuencia de aminoácidos SEQ ID Nº 11, una RHV-L2 que comprende la secuencia de aminoácidos SEQ ID Nº 12 y una RHV-L3 que comprende la secuencia de aminoácidos SEQ ID Nº 13. Reivindicación 20: Uno o más polinucleótidos aislados codificantes del inmunoconjugado según cualquiera de las reivindicaciones 1 a 19. Reivindicación 21: Uno o más vectores, particularmente vectores de expresión, que comprenden el polinucleótido o polinucleótidos según la reivindicación 20. Reivindicación 22: Célula huésped que comprende el polinucleótido o polinucleótidos según la reivindicación 20 o el vector o vectores según la reivindicación 21. Reivindicación 23: Método para producir un inmunoconjugado que comprende un polipéptido IL-2 mutante y un anticuerpo que se une a PD-1, que comprende: (a) cultivar la célula huésped según la reivindicación 21 bajo condiciones adecuadas para la expresión del inmunoconjugado y opcionalmente (b) recuperar el inmunoconjugado. Reivindicación 25: Composición farmacéutica que comprende el inmunoconjugado según cualquiera de las reivindicaciones 1 a 19 o 24 y un portador farmacéuticamente aceptable.
