Abstract
Изобретение описывает одностадийный способ получения конъюгата "связывающий клетку агент - цитотоксический агент", включающий приведение связывающего клетку агента в контакт с цитотоксическим агентом с образованием первой смеси, включающей связывающий клетку агент и цитотоксический агент, и приведение в контакт первой смеси, включающий связывающий клетку агент и цитотоксический агент, с бифункциональным реагентом, образующим перекрестные связи и включающем линкер, в растворе, имеющем pH от приблизительно 4 до приблизительно 9, с получением второй смеси, включающей конъюгат "связывающий клетку агент - цитотоксический агент", при этом связывающий клетку агент химически соединен посредством линкера с цитотоксическим агентом, несвязанный цитотоксический агент и побочные продукты реакции. Затем в некоторых случаях вторую смесь подвергают очистке с получением очищенного конъюгата "связывающий клетку агент - цитотоксический агент".
