Example 2
The invention provides a preparation method of a crushed wheat culture medium for cultivating cordyceps militaris, which comprises the following specific operations:
sieving wheat grains to remove impurities, and crushing the wheat grains by using a crusher with a sieve mesh aperture of 10 meshes at a set rotating speed of 420 rpm. Sieving the crushed wheat grains, and sieving and collecting the wheat grains with a grain size of-10 meshes to +30 meshes. The yield of the obtained wheat grains is 84.6%. Packing the wheat grains with a nylon-polyethylene composite plastic bag, wherein the packing thickness is 20cm, and performing irradiation modification treatment on the wheat grains by adopting an electron beam with the dose of 10kGy to obtain the crushed wheat. Mixing broken wheat with water according to the proportion of 1:2.0, stewing and sterilizing at 95 ℃ for 30min, cooling and forming to obtain the broken wheat culture medium for cultivating the cordyceps militaris, wherein the filling amount of the culture medium is 100g of broken wheat per box.
Diluting 10g/L Cordyceps militaris strain with sterile water under aseptic condition to obtain Cordyceps militaris liquid strain, and uniformly spraying Cordyceps militaris liquid strain onto the surface of the prepared crushed wheat culture medium, wherein each bottle is inoculated with 10mL of Cordyceps militaris liquid strain. Placing the inoculated cordyceps militaris culture medium into a culture room for dark culture, and culturing for 5 days at 18 ℃ until the surface of the cordyceps militaris culture medium is completely covered by cordyceps militaris mycelia; after completion of the dark culture, 1000Lux light was applied and cultivation management was performed at 18 ℃. Harvesting on 60 days after inoculation, drying the harvested Cordyceps militaris fruiting body at 60 deg.C for 24h, and calculating the output of Cordyceps militaris.
Example 3
The invention provides a preparation method of a crushed wheat culture medium for cultivating cordyceps militaris, which comprises the following specific operations:
sieving wheat grains to remove impurities, and crushing the wheat grains by using a crusher with a sieve mesh of 10 meshes at a set rotating speed of 460rpm. Sieving the crushed wheat grains, and sieving and collecting the wheat grains with a grain size of-10 meshes to +30 meshes. The yield of the obtained wheat grains is 95.0%. Packing the wheat grains with nylon-polyethylene composite plastic bags, wherein the packing thickness is 20cm, and performing irradiation modification treatment on the wheat grains by adopting gamma-rays with the dose of 2kGy to obtain the crushed wheat. Mixing broken wheat with water according to the proportion of 1:1.5, stewing and sterilizing at 100 ℃ for 40min, cooling and forming to obtain a broken wheat culture medium for cultivating cordyceps militaris, wherein the content of the culture medium is 100g of broken wheat per box.
Diluting 10 times of Cordyceps militaris strain with 10g/L sterile water under aseptic condition to obtain Cordyceps militaris liquid strain, uniformly spraying the Cordyceps militaris liquid strain onto the surface of the prepared crushed wheat culture medium, and inoculating 8mL of Cordyceps militaris liquid strain per bottle. Placing the inoculated cordyceps militaris culture medium into a culture room for dark culture, and culturing for 6 days at 18 ℃ until the surface of the cordyceps militaris culture medium is completely covered by cordyceps militaris mycelia; after completion of the dark culture, cultivation management was performed at 18 ℃ under illumination of 800Lux. Harvesting at 62 days after inoculation, drying the harvested Cordyceps militaris fruiting body at 60 deg.C for 24h, and calculating the output of Cordyceps militaris.
Comparative example 1
Sieving the complete wheat grains to remove impurities, and mixing the wheat grains with water according to the ratio of 1:1.8, stewing and sterilizing at 121 ℃ for 30min, cooling and forming to obtain the culture medium for cultivating the cordyceps militaris, wherein the loading amount of the culture medium is 100g of wheat grains/box.
Diluting 10 times of Cordyceps militaris strain with 10g/L sterile water under aseptic condition to obtain Cordyceps militaris liquid strain, uniformly spraying Cordyceps militaris liquid strain onto the surface of the prepared culture medium, and inoculating 8mL each bottle. Placing the inoculated cordyceps militaris culture medium into a culture room for dark culture, and culturing for 6 days at 18 ℃ until the surface of the cordyceps militaris culture medium is completely covered by cordyceps militaris mycelia; after completion of the dark culture, cultivation management was performed at 20 ℃ under illumination of 800Lux. Harvesting 55 days after inoculation, drying the harvested cordyceps militaris fruiting bodies at 60 ℃ for 24h, and calculating the output of cordyceps militaris.
Comparative example 2
Sieving the complete wheat grains to remove impurities, and mixing the wheat grains with water according to the ratio of 1:2.0, stewing and sterilizing at 123 ℃ for 30min, cooling and forming to obtain the culture medium for cultivating the cordyceps militaris, wherein the content of the culture medium is 100g of wheat grains/box.
Diluting 10g/L Cordyceps militaris strain with sterile water under aseptic condition to obtain Cordyceps militaris liquid strain, and uniformly spraying Cordyceps militaris liquid strain onto the surface of the prepared crushed wheat culture medium, wherein each bottle is inoculated with 10mL of Cordyceps militaris liquid strain. Placing the inoculated cordyceps militaris culture medium into a culture room for dark culture, and culturing for 5 days at 18 ℃ until the surface of the cordyceps militaris culture medium is completely covered by cordyceps militaris mycelia; after completion of the dark culture, 1000Lux light was applied and cultivation management was performed at 18 ℃. Harvesting on 60 days after inoculation, drying the harvested cordyceps militaris fruiting bodies at 60 ℃ for 24h, and calculating the output of cordyceps militaris.
Comparative example 3
Sieving complete wheat grains to remove impurities, and mixing the wheat grains with water according to the weight ratio of 1:1.5, steaming and sterilizing at 122 ℃ for 30min, cooling and forming to obtain the culture medium for cultivating the cordyceps militaris, wherein the loading amount of the culture medium is 100g of wheat grains/box.
Diluting 10g/L Cordyceps militaris strain with sterile water under aseptic condition to obtain Cordyceps militaris liquid strain, spraying Cordyceps militaris liquid strain onto the surface of the prepared crushed wheat culture medium, and inoculating 8mL of Cordyceps militaris liquid strain per bottle. Placing the inoculated cordyceps militaris culture medium into a culture room for dark culture, and culturing for 6 days at 18 ℃ until the surface of the cordyceps militaris culture medium is completely covered by cordyceps militaris mycelia; after completion of the dark culture, cultivation management was performed at 18 ℃ under illumination of 800Lux. Harvesting at 62 days after inoculation, drying the harvested Cordyceps militaris fruiting body at 60 deg.C for 24h, and calculating the output of Cordyceps militaris.
The unit yield of cordyceps militaris in each example and comparative example is counted, and the result is shown in table 1.

As can be seen from Table 1, the single-yield of the Cordyceps militaris can be increased by more than 36% by crushing the wheat grains, performing irradiation modification treatment, and reducing the sterilization temperature to prepare the culture medium, thereby significantly increasing the yield of the Cordyceps militaris.
Comparing the culture medium, the cordyceps militaris spawn running effect and the growth state in the embodiment 1 with those in the comparative example 1, as shown in fig. 1-3, it can be seen that the culture medium prepared by the method of the invention can enable cordyceps militaris spawn running to be denser, the growth speed to be faster and the number of sporophores to be larger.
The foregoing is only a preferred embodiment of the present invention, and it should be noted that, for those skilled in the art, various modifications and decorations can be made without departing from the principle of the present invention, and these modifications and decorations should also be regarded as the protection scope of the present invention.
Claims (10)
1. A preparation method of a crushed wheat culture medium for cultivating cordyceps militaris is characterized by comprising the following steps:
(1) Crushing wheat grains, and performing irradiation modification treatment to obtain crushed wheat;
(2) And mixing the crushed wheat with water, and performing cooking sterilization to obtain the crushed wheat culture medium for cultivating the cordyceps militaris.
2. The preparation method of claim 1, wherein the crushed wheat grains have a mesh size of-10 meshes to +30 meshes.
3. The method according to claim 2, wherein the irradiation is performed by electron beam irradiation or gamma-ray irradiation.
4. The production method according to claim 3, wherein the irradiation dose is 2 to 10kGy.
5. The preparation method according to claim 4, wherein during the irradiation, the crushed wheat grains are packaged by a nylon-polyethylene composite plastic bag, and the packaging thickness is 15-30 cm.
6. The method according to claim 5, wherein the mass ratio of the crushed wheat to the water is 1:1.4 to 2.0.
7. The method according to claim 6, wherein the temperature for the retort sterilization is 85 to 100 ℃.
8. The method of claim 7, wherein the retort sterilization is performed for 30 to 60min.
9. A crushed wheat culture medium for cultivating cordyceps militaris, which is prepared by the preparation method of any one of claims 1 to 8.
10. An application of the crushed wheat culture medium for cultivating cordyceps militaris in claim 9 in improving the yield of cordyceps militaris.
