Hi GO,
I would like to create a new term for pyroptotic cell death and modify modify the hierarchy and definition of of GO:0070269 pyroptosis.
The current problem is the following.
It is well-known that gasdermins trigger pyroptosis, which is commonly defined as a form of cell death. However, gasdermins form large pore that mediate release of mature proteins, such as IL1B and IL18), and do not always trigger programed-cell death (PMID:33883744).
In some, but not all, cells types, pyroptosis is followed by pyroptotic cell death, which is caused by downstream activation of ninjurin-1 (NINJ1), which mediates membrane rupture (cytolysis).
In other words, if pyroptosis is frequently associated with programmed cell- death, it is not always the case.
I prefer to create a new term related to cell death and change the hierarchy of pyroptosis because most papers usually test cell death in conditions that do not reflect in vivo conditions, and it is therefore almost impossible to assess whether cell death is relevant or not. By changing the hierarchy of existing annotations, we will therefore be sure that annotations are correct.
I will of course annotate "[pyroptotic cell death", but only in publications where we are certain that it leads to cell death.
