Abstract
1. Аутологичная вакцина для лечения онкологических заболеваний, содержащая лимфоциты, активированные интерлейкином 2, дендритные клетки, полученные путем инкубирования незрелых ДК с лизатом опухоли, отличающаяся тем, что содержит Т-лимфоциты специфически активированные зрелыми ДК. ! 2. Способ получения аутологичной вакцины для лечения онкологических заболеваний, включающий выделение мононуклеаров (МНК) из периферической крови пациента, культивирование МНК в среде DMEM, разделение клеток на моноциты, прикрепляющиеся к подложке и лимфоциты, не прикрепляющиеся к подложке, помещение МНК в культуральную среду, отделение не прикрепляющихся лимфоцитов и добавление к ним ИЛ2 для получения лимфокинактивированных киллеров (ЛАК-клеток, добавление к оставшимся прикрепляющимся моноцитам ростового фактора, стимуляцию созревания ДК аутологичным опухолевым лизатом in vitro и последующим добавлением созревающих факторов в течение 1 суток, отличающийся тем, что для получения прикрепляющихся моноцитов, МНК культивируют в среде, дополненной FCS 10% в течение одного часа, после чего, для получения незрелых дендритных клеток (нДК) проводят культивацию прикрепляющихся моноцитов в среде, дополненной нейпогеном 50 нг/мл в течение 48 ч, после чего, для получения зрелых дендритных клеток нДК культивируют в среде, содержащей 2000 МЕ/мл реоферона и 50 нг/мл беталейкина, при этом, параллельно культивируют ЛАК-клетки из неприкрепляющейся фракции МНК в среде содержащей 100 ЕД/мл ронколейкина в течение 72 ч, и, далее, полученные зДК и ЛАК-клетки отмывают центрифугированием в физиологическом растворе 10 мин при 1500 об/мин, после чего проводят совместную к1.
