Abstract
PURPOSE: To prepare tryptophan with a high productivity, by using coliform bacilli having an incorporated plasmid having a tryptophan operon released from the feedback inhibition by the tryptophan.
CONSTITUTION: Escherichia coil W3110 trp AE1 which is a strain of coliform bacilli derived from a strain of Escherichia coil K-12 subjected to the variant treatment, e.g. UV irradiation, and bacilli resistant to tryptophan among them are selected to give a strain in which the A Sase activity will not be inhibited by the tryptophan. The strain is then extracted to afford a recombined plasmid having a tryptophan operon released from the feedback inhibition by the tryptophan. Coliform bacilli in which the feedback inhibition is released by the introduction of the plasmid and the tryptophan repressor and the tryptophanase are deficient are cultivated in a culture medium, particularly containing anthranilic acid, and the tryptophan is then collected from the resultant culture.
