Abstract
PURPOSE: After brought into contact with a SH-group modifying reagent, γ-glutamyl-cystein synthetase is made to act on L-glutamic acid, L-cystein and glycine together with glutathione synthetase or produce the titled substance in high yield.
CONSTITUTION: γ-Glutamyl-cysteine synthetase is suspended in a buffer solution and brought into contact with a SH group-modifying reagent such as p-chlromercuribenzoic acid, or iodoacetic acid amide by adding the reagent to the suspension. The resultant γ-glutamy-cysteine synthetase and glutathione synthetase are made to act on L-glutamic acid, L-cystein and glycine together in an aqueous solution. An antioxidant, a surfactant and other additives may be added to the reaction mixture. The isolation and purification of glutathione from the reaction mixture is effected by a usual method.
