Description
Claims (1)
- 【特許請求の範囲】 1.工程: (a)少なくとも1つの第1炭水化物含有成分を有機シアン化試薬で活性化し て活性化された炭水化物を形成し、そして (b)前記活性化された炭水化物を第2成分に直接的または間接的にカップリ ングして、免疫応答を刺激することができる免疫原性構築物を形成する、 を含んでなる免疫原性構築物を製造する方法。 2.前記有機シアン化試薬が1−シアノ−4−(ジメチルアミノ)−ピリジニ ウムテトラフルオロボレート、N−シアノトリエチル−アンモニウムテトラフル オロボレート、またはp−ニトロフェニルシアネートである、請求項1に記載の 方法。 3.前記有機シアン化試薬が1−シアノ−4−(ジメチルアミノ)−ピリジニ ウムテトラフルオロボレートである、請求項1に記載の方法。 4.前記第1成分および第2成分が水溶性である、請求項3に記載の方法。 5.前記活性化工程(a)を8〜10のpHにおいて実施し、そして前記カッ プリング工程(b)を7〜9のpHにおいて実施する、請求項3に記載の方法。 6.前記活性化工程(a)をトリエチルアミンの存在において実施する、請求 項3に記載の方法。 7.第1成分を2官能性またはヘテロ官能性のスペーサー試薬に共有結合的に 結合し、かつ第2成分をスペーサー試薬に共有結合的に結合することによって工 程(b)を間接的に実施する、請求項1に記載の方法。 8.前記スペーサー試薬がエチレンジアミン、1,6−ヘキサンジアミン、ア ジピン酸ジヒドラジド、シスタミン、グリシン、およびリジンから成る群より選 択される、請求項7に記載の方法。 9.第1成分が多糖であり、そして第2成分がタンパク質である、請求項1に 記載の方法。 10.多糖がデキストラン、肺炎球菌(Pneumococcal)多糖、イン フルエンザ菌(Haemophilus influenzae)多糖、グルー プAストレプトコッカス(streptococcus)多糖、グループBスト レプトコッカス(streptococcus)多糖、および髄膜炎菌(N.m eningitidis)多糖から成る群より選択される、請求項9に記載の方 法。 11.第1成分が水溶性のウイルスまたは細菌の多糖である、請求項1に記載の 方法。 12.第2成分が水溶性タンパク質である、請求項1に記載の方法。 13.第2成分がウシ血清アルブミン、百日咳トキソイド、破傷風トキソイド、 マラリア誘導ペプチド、抗体、トキソイド、およびリポタンパク質から成る群よ り選択される、請求項1に記載の方法。 14.免疫原性構築物がPT−Pn、PT−PRP、TT−Pn、抗体−デキス トラン、およびペプチド−TT−Pnから成る群より選択される複合体である、 請求項1に記載の方法。 15.十分に低いpKaを有する求核物質を形成するように第2成分を誘導化す る、請求項1に記載の方法。 16.前記求核物質がヒドラジドである、請求項15に記載の方法。 17.前記カップリング工程(b)を約8より低いpHにおいて実 施する、請求項16に記載の方法。 18.十分に低いpKaを有する求核物質を形成するように第2成分を誘導化す る、請求項7に記載の方法。 19.前記求核物質がヒドラジドである、請求項18に記載の方法。 20.前記カップリング工程(b)を約8より低いpHにおいて実施する、請求 項19に記載の方法。 21.(a)(i)少なくとも1つの第1炭水化物含有成分を有機シアン化試薬 で活性化し、そして(ii)前記活性化された炭水化物を第2成分に共有結合的 に結合することを包含する工程により免疫原性構築物を調製し、そして (b)前記免疫原性構築物を患者に投与する、 ことを含んでなる、免疫応答を発生させる方法。 22.前記有機シアン化試薬が1−シアノ−4−(ジメチルアミノ)−ピリジニ ウムテトラフルオロボレートである、請求項21に記載の方法。 23.前記活性化工程(a)をトリエチルアミンの存在において実施する、請求 項22に記載の方法。 24.第1成分が多糖であり、そして第2成分が水溶性タンパク質である、請求 項22に記載の方法。 25.多糖がデキストラン、肺炎球菌(Pneumococcal)多糖、イン フルエンザ菌(Haemophilus influenzae)多糖、グルー プAストレプトコッカス(streptococcus)多糖、グループBスト レプトコッカス(streptococcus)多糖、および髄膜炎菌(N.m eningitidis)多糖から成る群より選択される、請求項24に記載の方 法。 26.第2成分がウシ血清アルブミン、百日咳トキソイド、破傷風トキソイド、 マラリア誘導ペプチド、抗体、トキソイド、およびリポタンパク質から成る群よ り選択される、請求項22に記載の方法。 27.免疫原性構築物がPT−Pn、PT−PRP、TT−Pn、抗体−デキス トラン、およびペプチド−TT−Pnから成る群より選択される複合体である、 請求項22に記載の方法。 28.十分に低いpKaを有する求核物質を形成するように第2成分を誘導化す る、請求項21に記載の方法。 29.前記求核物質がヒドラジドである、請求項28に記載の方法。 30.前記カップリング工程(b)を約8より低いpHにおいて実施する、請求 項29記載の方法。
