Description
本願は、2006年12月21日に出願された米国仮出願第60/876,801号(その開示の全体が、参照により、本明細書に組み込まれる。)の利益を主張する。
本願は、全般的には、疾病を治療するための医薬、例えば、生物分子医薬又は生物医薬のための製剤に関する。
抗体の安定性に対する様々な製剤の効果を測定するために、下表2に示されている幾つかの製剤を調製した。製剤化された抗体溶液約3.0mLを、様々な時間にわたって、異なる温度で含有する、ゴム製Daikyoフッ素重合体栓付きの5ccブローバック型タイプ1ガラスバイアル中に製剤を保存した。
SECを用いて、一般に、E51P30製剤は、保存時間にわたって、主ピークのパーセントの最も少ない喪失を示すが、幾つかの製剤が特定の期間にわたって同等の結果を示し得る。4℃で12ヶ月又は18ヶ月間(図1A)、25℃で8週間、13週間又は25℃で6ヶ月(図1B)、特に37℃で6ヶ月(図1C)の保存後に、保存時間にわたっての主ピークのパーセントのより少ない低下が明瞭である。しかしながら、E51T30及びE51M30製剤も、ある温度で、増加した安定性を提供し得ることに注目すべきである。
バイアル又は予め充填された注射器中での保存中の様々な製剤の安定性を調べた。37℃で、1週、2週、1ヶ月及び2ヶ月後に、抗体を含む製剤の安定性を調べた。異なる温度で、様々な時間にわたって、バイアル又は予め充填された注射器の中に製剤を保存した。表3は、この研究で用いた幾つかの異なる製剤を列記している。
Claims (13)
- 4.5〜5.5のpHを有する5〜50mMのグルタミン酸緩衝液、2%〜10%の濃度のプロリン、及び抗神経成長因子(NGF)抗体又はその抗原結合断片を含む製剤であって、前記抗NGF抗体又はその抗原結合断片の濃度が3〜100mg/mLである、製剤。
- グルタミン酸緩衝液が10mM、30mM又は50mMの濃度、及び5のpHを含む、請求項1に記載の製剤。
- 等張濃度を有する、請求項1に記載の製剤。
- 前記抗体又は抗原結合断片が10〜50mg/mLの濃度を有する、請求項1に記載の製剤。
- グルタミン酸緩衝液がグルタミン酸ナトリウムから調製される、請求項1に記載の製剤。
- pHが5.0、5.1又は5.2である、請求項1に記載の製剤。
- 4.5〜5.5のpHを有する5〜50mMのグルタミン酸緩衝液、2%〜10%のプロリン、及び抗神経成長因子(NGF)抗体又はその抗原結合断片の3〜100mg/mlを組み合わせることを含む、製剤を調製する方法。
- 4.5〜5.5のpHを有する3〜50mMのグルタミン酸、2%〜10%のプロリン、及び抗神経成長因子(NGF)抗体又はその抗原結合断片を有する水溶液を含む製剤、を含有する容器であって、抗NGF抗体又はその抗原結合断片の濃度が3〜100mg/mLである、容器。
- 容器がバイアル又は予め充填された注射器である、請求項8に記載の容器。
- 患者における神経成長因子の増加した発現又は神経成長因子に対する増加した感受性によって引き起こされる症状を治療するための請求項1に記載の製剤。
- 症状が疼痛又は神経因性疼痛である、請求項10に記載の製剤。
- 4.5〜5.5のpHを有する5〜50mMのグルタミン酸緩衝液又はこのような緩衝液を調製するための適切なグルタミン酸塩と、2%〜10%の濃度のプロリンと、抗NGF抗体又はその抗原結合断片と、並びにこれらの使用に関する指示書と、を1つ以上の容器中に含むキットであって、抗NGF抗体または抗原結合部位の濃度が3〜100mg/mLである、キット。
- 4.5〜5.5のpHを有する5〜50mMのグルタミン酸緩衝液又はこのような緩衝液を調製するための適切なグルタミン酸塩と、2%〜10%の濃度のプロリンと、一緒に混合したときに、混合物の保存の間、プロリン及び抗NGF抗体又はその抗原結合断片が4.5〜5.5のpHを維持する濃度の抗NGF抗体又はその抗原結合断片と、並びにこれらの使用に関する指示書とを1つ以上の容器中に含むキットであって、抗NGF抗体又はその抗原結合断片の濃度が3〜100mg/mLである、キット。
