Abstract
Description

RPMI1640に、25mMのHEPESと、1mMのピルビン酸ナトリウムと、0.1mMの非必須アミノ酸と、1mMのL−グルタミン(ミネソタ州ミネアポリスのセロックス・ラボラトリーズ,インコーポレーティッド(Celox Laboratories、Inc.,Minneapolis,MN))とを混合し、10%の加熱不活性化したウシ胎児血清(FCS)(ユタ州ローガンのハイクローン・ラボラトリーズ、インコーポレーティッド(Hyclone Laboratories,Inc.,Logan,UT))と1%のペニシリン/ストレプトマイシン(ミズーリ州セントルイスのシグマ・ケミカル・カンパニー(Sigma Chemical Co.,St Louis,MO))を補足して、完全なRPMIを調製した。
2mMのLグルタミンと、1.5g/Lの炭酸水素ナトリウム、0.1mMの非必須アミノ酸および1.0mMのピルビン酸ナトリウムを含有するよう調製したアールの緩衝塩類溶液(Earle’s Balanced Salt Solution)(メリーランド州ロックビルのインビトロゲン・コーポレーション(Invitrogen Corp.,Rockville,MD))を含有する90%の最小必須培地(Minimum Essential Medium)(MEM)と10%の加熱不活性化ウシ胎児血清の中で、HEK293細胞を培養した。細胞は、37℃、8%CO2でインキュベートした。
pH8.0の0.5MのEDTA(ニューヨーク州グランドアイランドのギブコ(Gibco,Grand Island,NY))750μLが入った60mLシリンジに全血を採取した。血液を、カルシウムまたはマグネシウムを含まないダルベッコ燐酸緩衝生理食塩水(Dulbecco’s Phosphate Buffered Saline)(DPBS、カリフォルニア州カマリロのバイオソース・インターナショナル(Biosource International,Camarillo,CA))で1:1の希釈を行い、ヒストパクー1077(Histopaque−1077)(ミズーリ州セントルイスのシグマ・ケミカル・カンパニー(Sigma Chemical Co.,St Louis,MO))を被せた。細胞を、25℃、2000RPMで30分間遠心分離した。軟膜層を分離し、25℃、1350RPMで10分間、DPBSにより3回洗浄した。

実施例1に記載したようにしてHEK293細胞への導入を行った。HEK293培養細胞を、濃度0.1μM、0.3μM、1.0μM、3.0μM、10μM、30μM、または、ビヒクルコントロールの、IRM化合物または陰性コントロール化合物(1,5−ジヒドロ−1−(2−メチルプロピル)−4H−イミダゾ[4,5−c]キノリン−4−オン、米国特許第4,698,348号明細書、実施例71)と共にインキュベートした。培養細胞を、その他の点では実施例1に記載したようにしてインキュベートした。ルシフェラーゼシグナルを、実施例1に記載したようにしてRLU(相対ルシフェラーゼ単位)で読み取った。結果を図7〜図11および表3に示す。
Claims (29)
- TLR8発現細胞内でTLR8介在細胞応答を引き出す方法であって、以下のステップ:
TLR8アゴニストとして同定された化合物を選択すること;および
少なくとも1つのTLR8介在細胞シグナル伝達経路に影響を与える量の前記化合物を前記細胞に投与すること;
を含み、
ここで、前記TLR8アゴニストが、置換イミダゾキノリンアミン;テトラヒドロイミダゾキノリンアミン;イミダゾピリジンアミン;1,2−架橋イミダゾキノリンアミン;6,7−縮合シクロアルキルイミダゾピリジンアミン;イミダゾナフチリジンアミン;テトラヒドロイミダゾナフチリジンアミン;オキサゾロキノリンアミン;チアゾロキノリンアミン;オキサゾロピリジンアミン;チアゾロピリジンアミン;オキサゾロナフチリジンアミン;チアゾロナフチリジンアミン;6−,7−,8−もしくは9−アリールまたはヘテロアリール置換イミダゾキノリンアミン;あるいは、ピリジンアミン、キノリンアミン、テトラヒドロキノリンアミン、ナフチリジンアミンまたはテトラヒドロナフチリジンアミンと縮合した1H−イミダゾダイマーである、前記方法。 - 前記細胞が、単球、マクロファージ、樹状細胞、Bリンパ球、ナチュラルキラー細胞、多形核細胞またはこれらから誘導される細胞である、請求項1に記載の方法。
- 前記細胞応答が、NF−κBの活性化、少なくとも1つのサイトカインの産生、少なくとも1つの共刺激マーカーの産生またはこれらのあらゆる組み合わせのいずれかを含む、請求項1に記載の方法。
- TLR8介在細胞応答を調節することによって治療可能な状態を有する生体を治療する方法であって、以下のステップ:
TLR8アゴニストとして同定された化合物を選択すること;および
TLR8介在細胞シグナル伝達経路を調節するのに有効な量の前記化合物を前記生体に投与すること;
を含み、
ここで、前記TLR8アゴニストが、置換イミダゾキノリンアミン;テトラヒドロイミダゾキノリンアミン;イミダゾピリジンアミン;1,2−架橋イミダゾキノリンアミン;6,7−縮合シクロアルキルイミダゾピリジンアミン;イミダゾナフチリジンアミン;テトラヒドロイミダゾナフチリジンアミン;オキサゾロキノリンアミン;チアゾロキノリンアミン;オキサゾロピリジンアミン;チアゾロピリジンアミン;オキサゾロナフチリジンアミン;チアゾロナフチリジンアミン;6−,7−,8−もしくは9−アリールまたはヘテロアリール置換イミダゾキノリンアミン;あるいは、ピリジンアミン、キノリンアミン、テトラヒドロキノリンアミン、ナフチリジンアミンまたはテトラヒドロナフチリジンアミンと縮合した1H−イミダゾダイマーである、前記方法。 - 前記生体が哺乳動物である、請求項4に記載の方法。
- 前記哺乳動物がヒトである、請求項5に記載の方法。
- 前記状態が新生物形成疾患である、請求項6に記載の方法。
- 前記状態がTH2介在疾患である、請求項6に記載の方法。
- 前記状態が、喘息、アレルギー性鼻炎またはアトピー性皮膚炎である、請求項8に記載の方法。
- 前記状態が、ウイルス性疾患、細菌性疾患、寄生虫病、原虫病またはプリオン介在疾患である、請求項6に記載の方法。
- 前記IRM化合物が、NF−κB活性、少なくとも1つのサイトカインの産生、少なくとも1つの共刺激マーカーの産生、細胞間接着分子の産生、成熟マーカーの産生またはこれらのあらゆる組み合わせのいずれかを調節する、請求項4に記載の方法。
- TLR8アゴニストを同定する方法であって、以下のステップ:
a)TLR8陽性細胞培養を試験化合物に曝露し、TLR8介在細胞応答を測定すること;
b)TLR8陰性細胞培養を試験化合物に曝露し、TLR8介在細胞応答を測定すること;および
c)前記TLR8陽性細胞培養の細胞応答が前記TLR8陰性細胞培養の細胞応答より大きいとき、上記試験化合物をTLR8アゴニストとして同定すること;
を含む、前記方法。 - 前記TLR8陰性細胞培養が、TLR8のドミナントネガティブ変異体を発現する細胞を含む、請求項12に記載の方法。
- 前記TLR8陰性細胞培養が、TLR8に対して産生された抗体を含む、請求項12に記載の方法。
- 前記TLR8陽性細胞培養が、TLR8を過剰発現する細胞を含む、請求項12に記載の方法。
- 前記TLR8陽性細胞培養の細胞応答が前記TLR8陰性細胞培養の細胞応答より少なくとも20%大きいとき、上記試験化合物をTLR8アゴニストとして同定する、請求項12に記載の方法。
- 前記TLR8陽性細胞培養の細胞応答が前記TLR8陰性細胞培養の細胞応答より少なくとも50%大きいとき、上記試験化合物をTLR8アゴニストとして同定する、請求項12に記載の方法。
- 前記TLR8陽性細胞培養の細胞応答が前記TLR8陰性細胞培養の細胞応答より少なくとも80%大きいとき、上記試験化合物をTLR8アゴニストとして同定する、請求項12に記載の方法。
- 前記TLR8介在細胞応答が、NF−κBの活性化、少なくとも1つのサイトカインの産生、少なくとも1つの共刺激マーカーの産生またはこれらのあらゆる組み合わせのいずれかを含む、請求項12に記載の方法。
- 請求項12に記載の方法によりTLR8アゴニストとして同定された化合物。
- 薬学的に受容可能な担体と組み合わされたTLR8アゴニストを含む医薬組成物。
- TLR8アンタゴニストを同定する方法であって、以下のステップ:
a)第1のIRM応答性細胞培養をTLR8アゴニストに曝露し、TLR8介在細胞応答を測定すること;
b)第2のIRM応答性細胞培養をTLR8アゴニストおよび試験化合物に曝露し、TLR8介在細胞応答を測定すること;および
c)前記第1の細胞培養の細胞応答が前記第2の細胞培養の細胞応答より大きいとき、上記試験化合物をTLR8アンタゴニストとして同定すること;
を含む、前記方法。 - 前記TLR8アゴニストが、置換イミダゾキノリンアミン;テトラヒドロイミダゾキノリンアミン;イミダゾピリジンアミン;1,2−架橋イミダゾキノリンアミン;6,7−縮合シクロアルキルイミダゾピリジンアミン;イミダゾナフチリジンアミン;テトラヒドロイミダゾナフチリジンアミン;オキサゾロキノリンアミン;チアゾロキノリンアミン;オキサゾロピリジンアミン;チアゾロピリジンアミン;オキサゾロナフチリジンアミン;チアゾロナフチリジンアミン;6−,7−,8−もしくは9−アリールまたはヘテロアリール置換イミダゾキノリンアミン;あるいは、ピリジンアミン、キノリンアミン、テトラヒドロキノリンアミン、ナフチリジンアミンまたはテトラヒドロナフチリジンアミンと縮合した1H−イミダゾダイマーである、請求項22に記載の方法。
- 請求項22に記載の方法によりTLR8アンタゴニストとして同定された化合物。
- 薬学的に受容可能な担体と組み合わされたTLR8アンタゴニストを含む医薬組成物。
- TLR8介在細胞シグナル伝達経路を活性化させる化合物を同定するためのTLR8のドミナントネガティブ変異体の使用。
- TLR8の活性化を検知する分析評価法におけるIRM化合物の陽性コントロールとしての使用であって、前記IRM化合物が、置換イミダゾキノリンアミン;テトラヒドロイミダゾキノリンアミン;イミダゾナフチリジンアミン;テトラヒドロイミダゾナフチリジンアミン;オキサゾロキノリンアミン;チアゾロキノリンアミン;オキサゾロピリジンアミン;チアゾロピリジンアミン;オキサゾロナフチリジンアミン;チアゾロナフチリジンアミン;6−,7−,8−もしくは9−アリールまたはヘテロアリール置換イミダゾキノリンアミン;あるいは、ピリジンアミン、キノリンアミン、テトラヒドロキノリンアミン、ナフチリジンアミンまたはテトラヒドロナフチリジンアミンと縮合した1H−イミダゾダイマーを含む、前記使用。
