Abstract
Non-toxic acid addition salts of N-(1-pyrrolidinylmethyl) tetracycline are prepared by reacting the tetracycline base with the appropriate acid. Specified salts are the hydrochloride, sulphate, sulphamate, tartrate, hydrobromate, hydriodate, glycolate, citrate, maleate, phosphate, polyimido-phosphate, succinate, acetate and ascorbate. Specifications 809,585 and 817,180 are referred to.ALSO:A substantially anhydrous therapeutic composition for intramuscular injection when dissolved in sterile water or saline, comprises N-(1-pyrrolidinylmethyl) tetracycline, or a non-toxic salt thereof, and a non-toxic acidifying agent in a sufficient amount to produce a pH of not more than 4 when the mixture is dissolved for use. The acidifying agent may be (a) an a -hydroxy-carboxylic acid containing no elements other than carbon, hydrogen and oxygen, e.g. citric, gluconic, tartaric, maleic, malic, ascorbic, mandelic and pyruvic acids; (b) carbamide phosphoric acid; (c) glycine hydrochloride; (d) glutamic acid hydrochloride or (e) sulphamic acid. Suitable salts of N - (1 - pyrrolidinylmethyl) tetracycline are the hydrochloride, sulphate, sulphamate, tartrate, hydrobromate, hydriodate, glycolate, citrate, maleate, phosphate, polyimidophosphate, succinate, acetate and ascorbate. A local anaesthetic such as procaine hydrochloride and lignocaine hydrochloride may also be present. Mixtures providing a pH of 3 to 4 and especially pH 3.4 are preferred. The preparations provide high blood levels of tetracycline. Specifications 809,585 and 817,180 are referred to.
