Abstract

12a-hydroxylation of a 12a-deoxytetracycline is obtained by treating the 12a-deoxytetracycline at a pH of 2 to 7 with an inorganic oxidizing agent effective to cause 12a-hydroxylation under aerobic conditions. Specified preferred oxidizing agents are sodium nitrite, potassium nitrite, potassium permanganate, potassium dichromate, potassium ferricyanide, potassium iodide, iodine and potassium persulphate, but sodium hydrochlorite or potassium iodate may also be used. Typical 12a-deoxytetracyclines which may be hydroxylated are 12a-deoxytetracycline, 7-chloro-6-dimethyl-12a-deoxytetracycline, 6,12a-di-deoxytetracycline, 6 - demethyl - 12a - deoxytetracycline, 6 - demethyl - 6,12a - dideoxytetracycline and 12a-deoxydedimethyl-aminotetracycline.
