Abstract
1,267,398. 5α,6α-Epoxysteroids. SCHERING A.G. 14 April, 1969 [13 April, 1968], No. 19009/69. Heading C2U. The invention firstly comprises the preparation of 3#,11#-dihydroxy-5α,6α-epoxy steroids by enzymatic 11-hydroxylation of 3#-(hydroxy or acyloxy)-1 11-deoxy-5α,6α-epoxy steroids using Curvularia, e.g. C. lunata. Any initial acyloxy group in the 3-position or elsewhere is hydrolysed concurrently. In Example 4, the 3- and 21-hydroxy groups of the reaction product are subsequently acetylated. The invention secondly comprises the following products: 3#,11#-dihydroxy-5α,6α-epoxypregnan-20-one and the 17α-hydroxy, 17α,21- dihydroxy, 16α-methyl, 21-hydroxy-16α-methyl, 17α,21 - dihydroxy - 16α - methyl, 16α,17α- epoxy and 16α,17α-epoxy-16#-methyl derivatives thereof; 11#-hydroxy-3#,21-diacetoxy-5α, 6α - epoxy - 16α - methylpregnan - 20 - one; 3#,- 11#,20# - trihydroxy - 5α,6α - epoxypregnane.; 3#,11#,17# - trihydroxy - 5α,6α - epoxy - 17α- pregnan - 20 - one; 3#,11#,21 - trihydroxy - 5α,- 6α,17α,20 - diepoxypregnane; 3#,11#,17# - trihydroxy-5α,6α-epoxyandrostane and its 17α- methyl derivative; and 3#,11#,17α-trihydroxy- 5α,6α-epoxyandrostane. Other products disclosed are 3#,11#,21-trihydroxy - 5α,6α - epoxypregnan - 20 - one (Ex. 2) and 3#,11#-dihydroxy-5α,6α-epoxyandrostan- 17-one (Ex. 7). The 3# (hydroxy or acyloxy - 11 -deoxy- 5α,- 6α-epoxy starting materials are prepared by epoxidation of the corresponding #<SP>5</SP>-compounds.
