Description


  



   L'invenzione qui descritta si riferisce a delle composizioni farmaceutiche,  a l'utilizzazione di composti  come agenti  ad attività promitrice di assorbimento di  principi  attivi ed a composti  specifici, tutto secondo le parti generali delle rivendicazione 1, 2, 5, 6 e 10 seguenti. 



  Problemi legati all'assorbimento cutaneo dei p.a. in forme farmaceutiche topiche e transdermali (drug delivery systems) è noto ed oggetto di brevettazione [1]. Percutaneous absorption: mechanisms-methodology-drug delivery, 2 Ed., Bronaugh Maibach, MARCEL DEKKER, INC. 



  Tra i più significativi esempi riportati nella letteratura scientifica possiamo menzionare l'attività enhancer del decilmetilsolfossido per incrementare la permeazione cutanea dell'idoxuridina [2] Touitou, E. (1988). lnt. J. Pharm. 43:1.  L'effetto del dodecilazacicloeptan-2-one (laurocapram  Azone)  di promuovere l'assorbimento di varie molecole è stato indagato da Stoughton [3] Stoughton, R.B. (1982) Arch. Dermatol. 118:474. e da Sugibayashi [4] Sugibayashi, K., Hosoya, K., Morimoto,Y., and Higuchi, W. I. (1985). J. Pharm. Pharmacol. 37:578. 



  II mercato ha messo a disposizione sostanze inizialmente con un utilizzo in campi differenti dal farmaceutico che poi si sono rivelati di notevole importanza nel campo farmaceutico. Tra queste, possiamo trovare una classe di composti Perfluoropolieteri (PFPE)  [5] ottenuti dalla polimerizzazione di esafluoropropene con ossigeno attivato dalle radiazioni UV e successivamente trattato per ottenere un prodotto di grado cosmetico [6] quale eccipiente per preparati atti alla preparazione i prodotti ad effetto barriera [7]. 


 Riassunto dell'invenzione 
 



  La  seguente invenzione consiste nell'utilizzare i  PFPE quali promotori di  assorbimento di  PA farmacologicamente attivi  in forme farmaceutiche topiche e/o in formulazioni transdermali (drug delivery systems) contenenti miscele 0 almeno un PA per promuoverne l'assorbimento. 



  Questi composti se  inclusi in formulazioni farmaceutiche contenenti una associazione o almeno un PA possono modificare la capacità di permeazione dei PA attraverso la cute, evidenziando una sorprendente e inaspettata capacità di promuovere la penetrazione dei PA. 



  In accordo con il primo aspetto dell'invenzione, l'inclusione di una quantità di PFPE può essere applicata in formulazioni topiche convenzionali, quali ad esempio, ma non esclusivamente: creme, emulsioni liquide, unguenti, lozioni, microemulsioni, schiume, gel, polveri aspersonrie; ed in formulazioni  transdermali (drug delivery systems). 



  Con il termine di PFPE si intende molecole con struttura chimica

  Con pesi molecolari compresi tra i 650 e 6250,

  È stato sorprendemente trovato che l'inclusione di una quantità di PFPE in concentrazioni comprese tra lo 0,01% p/p e il 60% p/p, in formulazioni topiche e/o transdermali (drug delivery systems) riesce ad incrementare la permeabilità cutanea di PA a oltre 5-20 volte la loro capacità naturale, permettendo cosÌ di migliorarne l'assorbimento e ottimizzarne l'effetto sistemico attraverso adatte forme farmaceutiche. 


 Descrizione dettagliata dell'invenzione 
 



  La comprensione dell'invenzione è facilitata dalla descrizione di alcune valutazioni della permeabilità cutanea di alcune formulazioni contenenti differenti PA. 



  Nell'esempio A viene evidenziato l'aumento della permeabilità di circa 10 volte di una formulazione a base di troxerutina contenente il 3% di PFPE rispetto alla formulazione base. 



  Nell'esempio B viene dimostrata la correlazione concentrazione PFPE-permeazione cutanea di formulazioni contenenti Nimesulide e concentrazioni crescenti di PFPE. 



  Nell'esempio C vengono comparate le velocità di permeazione di formulazioni contenenti alcuni antinfiammatori non steroidei versus formulazioni addizionate del 3% di PFPE. 


 Esempio A 
 



  Verifica del passaggio di TROXERUTINA attraverso cute porcina di differenti formulazioni contenenti diversi tipi di promotori di assorbimento. In particolare è stato studiato il profilo di rilascio della TROXERUTINA da formulazioni contenenti PFPE, Acido nor-chenodesossicolico e transcutol, verso una formulazione di riferimento. 



  Parte sperimentale: 



  Apparecchiature: Celle di Franz (produttore: Crown Glass Company Inc. New Jersey). Le celle di diffusione di Franz sono uno dei sistemi principali per gli studi di permeazione in ex-vivo. In questo studio si sono utilizzate tre celle aventi ciascuna un compartimento donatore di 9 mm di diametro corrispondente ad un'area di diffusione di 64 mm<2>.
 Il compartimento ricevente ha una capienza di 4,8 ml. La camera recettoriale è ermostatata a 37 DEG C +/-1.

  Preparazione della cute 



  I campioni di cute porcina sono stati prelevati dall'orecchiointerno di maiale appea sacrificato.
 I campioni vengono immersi per 24 ore a 4 DEG C in una soluzione di tampone a pH 7,4.
 Sucessivamente vengono asportate le parti connettivali e muscolari della cute, quindi viene sucessivamente tagliata in pezzi quadrati quadrati da 2 cm di lato.
 I campioni di cute vengono conservati ad una temperatura di -20 DEG C. 



  Condizioni operative di analisi HPLC 



  Colonna: KP18, 5 micron Licrospher 100
 Fase mobile: 20% acetonitrile 80% tampone fosfato a pH 6,6
 Flusso 1 ml/min
 Pressione 1,52 Kpsi
 Lunghezza d'onda: 245 nm
 TR: 8,51 



  Modalità di esecuzione 



  Si posiziona la cute porcina tra il compartimento donatore e la camera recettrice con la superficie cutanea esterna rivolta verso il compartimento superiore. Si inserisce la formulazione da testare all'interno del compartimento donatore.
 La camera ricevente viene riempita con soluzione tampone fosfato a pH 7,4, e ad intervalli di tempo compresi tra 0,5 ore e 8 ore, si prelevano dalla camera recettoriale 2 ml di soluzione. Ad ogni prelievo si ripristina il volume iniziale con soluzione tampone fresca, termostatata a 37 DEG C. I campioni vengono quindi analizzati mediante metodica HPLC. Ogni prova viene effettuata in triplicato.

   Conclusioni:
 Le formulazioni B,C,D non hanno evidenziato alcuna differenza di permeabilità, mentre la formulazione A, contenente PFPE ha evidenziato all'ottava ora, una permeabilità di circa 9-10 volte maggiore della formulazione base D.

  Verifica del passaggio di NIMESULIDE mediante celle di Franz, come descritto nell'esempio A, attraverso cute porcina di differenti formulazioni contenenti diversi concentrazioni di PFPE verso una formulazione di riferimento.

  Conclusioni:
 Le formulazioni B,C,D hanno evidenziato crescenti capacità di permeare la cute, questa capacità permeante risulta essere correlata alla dose crescente di PFPE.

Claims (10)

1. Composizioni farmaceutiche, caratterizzate dal fatto di contenere, oltre il o i principio(i) attivo(i), - tra lo 0,01 ed il 60% p/p dei composti di formula I EMI22.1 - con n e m > 18 e < 46 e con pesi molecolari fra 600 e 8000- in combinazione con da 0,01 a 20% p/p di Fosfatidilcolina- come composti ad attività promotrice di assorbimento dei principi attivi.
2. Composizioni farmaceutiche secondo la rivendicazione 1 con 0,1 a 30% p/p dei composti di formula I con n e m > 24 e < 36 e con pesi molecolari fra 1000 e 4000 e con 0,1 a 10% p/p di Fosfatidilcolina.
3. Composizioni farmaceutiche secondo una delle rivendicazioni 1 o 2, contenenti altre componenti compatibili e presenti in forma di creme, emulsioni, unguenti, lozioni, schiume, gel e formulazioni trans-dermali.

Utilizzazione dei composti di formula I EMI23.1 con n e m > 18 e < 46, preferibiImente > 24 e < 36, e con pesi molecolari fra 600 e 8000, preferibilmente fra 1000 e 4000, congiuntemente con la Fosfatidilcolina, in composizioni farmaceutiche ad uso topico esterno o interno per promuovere l'assorbimento di prinicpi attivi attraverso derma, cutis, mucosa, rectum, vagina e urethra.

Utilizzazione secondo la rivendicazione 4, caratterizzata dal fatto che principi attivi usati hanno preferibilmente un effetto anabolico, analgesico, androgenico, anestetico, anoressico, antelmintico, antiallergico, antiamebico, antiandrogenico, anti-anginale, antiarritmico, antiarteriosclerotico, antiartritico e -reumatico, antibatterico, anticolinergico, anticonvulsivo, antidepressivo, antidiabetico, antidiarroico, antidiuretico, antiestrogenico, antibiotico, antiglaucomico, antigonatropico, antiistaminico, antiiperlipoproteico, antiipertireodico, antiipertensivo, antiinfiammatorico, antimalarico, antimigranico, antinauseante, antineoplastico, antiparkinsonico, antiprotozoico, antipruritico, antipsoriatico, antipsicotico, antipiretico, antisettico, antispasmodico, antitrombotico, antitussivo, antiulcerale, antivirale, ansiolitico, broncodilatatorio, CA-bloccante o -regolatorio,
cardiotonico, stimolante, decongestionante, diuretico o enzimatico.
6. Utilizzazione secondo la rivendicazione 5, caratterizzata dal fatto che i principi attivi sono la Troxerutina o il Nimesulide ed, inoltre, agenti antiinfiammatori non-steriodali come la Ketoprofene, il Diclofenac Sodio, 1¾Ibuprofene, l'acido Etodolico e il Piroxicam.
7. Utilizzazione secondo la rivendicazione 4, caratterizzata dal fatto che l'assorbimento dei farmaci è incrementato fino a oltre 5 volte la loro capacità naturale. EMI24.1
8. Composti della classe dei Perfluoropolieteri di formula I con n e m > 18 e < 46 e con pesi molecolari fra 600 e 8000.
9. Composti secondo la rivendicazione 8 con n e m > 24 e < 36 e con pesi molecolari fra 1000 e 4000.

Composti secondo la rivendicazione 9, caratterizzati dal fatto che sono ottenuti mediante polimerizzazione di esafluoropropene EMI24.2 con ossigeno attivato all'UV.
