Abstract
"DNA CODIFICANDO PARA UMA PROTEìNA, CéLULA, E, PROCESSO PARA PRODUZIR ISOMERASE DE FOSFATO DE HEXULOSE". A sintase de fosfato de hexulose foi purificado de <MU>Mycobacterium<MV> gastri_para determinar uma sequência de aminoácido da enzima. Os iniciadores de oligonucleotídeos a serem usados para PCR foram sintetizados em bases de informação da sequência de aminoácido obtida. Um fragmento de DNA, que foi amplificado por realização do PCR por uso de um gabarito de DNA genómico preparado a partir de <MU>Mycobacterium<MV> <MU>gastri<MV> foi usado como uma sonda de modo que a hibridização da colónia foi realizada com relação a uma coleção compreendendo fragmentos obtidos por digestão de DNA genómico de <MU>Mycobacterium<MV> <MU>gastri<MV> com <MU>Pst<MV>I para obter um clone positivo. Assim, o fragmento de DNA que contém ORF codificando sintase de fosfato de hexulose foi obtido. Além disso, o fragmento de DNA clonado continha outro ORF (ORF-2). O sistema de expressão de ORF-2 foi construído (pT-HPIS-1) e foi introduzido em célula de <MU>Escheichia coli<MV>. Após a indução da expressão de ORF-2, a atividade de sintase de fosfato de hexulose foi verificada no extrato de células abrigando pT-HPIS-1. Assim, o gene, que codificou para sintase de fosfato de hexulose, foi obtido junto com ORF que codificou para sintase de fosfato de hexulose."DNA CODING FOR A PROTEIN, CELL, AND PROCESS TO PRODUCE HEXULOSE PHOSPHATE ISOMERASE". The hexulose phosphate synthase was purified from <MU> Mycobacterium <MV> gastri_ to determine an amino acid sequence of the enzyme. The oligonucleotide primers to be used for PCR were synthesized in information bases of the obtained amino acid sequence. A DNA fragment, which was amplified by performing PCR using a genomic DNA template prepared from <MU> Mycobacterium <MV> <MU> gastri <MV> was used as a probe so that colony hybridization was performed with respect to a collection comprising fragments obtained by digesting genomic DNA from <MU> Mycobacterium <MV> <MU> gastri <MV> with <MU> Pst <MV> I to obtain a positive clone. Thus, the DNA fragment containing ORF encoding hexulose phosphate synthase was obtained. In addition, the cloned DNA fragment contained another ORF (ORF-2). The ORF-2 expression system was constructed (pT-HPIS-1) and was introduced into a <MU> Escheichia coli <MV> cell. After the induction of ORF-2 expression, hexulose phosphate synthase activity was verified in the cell extract harboring pT-HPIS-1. Thus, the gene, which encoded hexulose phosphate synthase, was obtained together with ORF which encoded hexulose phosphate synthase.
