Artificial Intelligence Predicts Features Associated with Breast Cancer Neoadjuvant Chemotherapy Response from Multi-stain Histopathologic Images

IRB Information

This study was approved by the Ohio State University Institutional Research Board and included 62 HER2-positive breast cancer (HER2+) patients and 64 triple-negative breast cancer (TNBC) patients treated with neoadjuvant chemotherapy (NAC) and follow-up surgical excision. Patients with histopathologically confirmed invasive breast carcinoma who underwent NAC from January 2011 to December 2016, those who had underwent surgery after completing NAC were included.

Workflow

We aimed to investigate whether artificial intelligence (AI)-based algorithms using automatic feature extraction methods can predict neoadjuvant chemotherapy (NAC) outcome in HER2-positive (HER2+) and triple-negative breast cancer (TNBC) patients. In this study, an automatic, accurate, comprehensive, interpretable, and reproducible whole slide image (WSI) feature extraction pipeline was constructed, IMage-based Pathological REgistration and Segmentation Statistics (IMPRESS). The developed machine learning models using image-based features derived from tumor and tumor microenvironment, biomarkers and clinical features accurately predicted the response to NAC in breast cancer patients, and outperformed the results learned by pathologists' manually assessed features.

Run Experiments

  • Users can use main.py to run machine learning experiments.

  • After machine learning experiments, users can use summary.py to generate results.

  • Alternatively, a bash file can help users to run all experiments & analyses altogether using 10 different random seeds:
