Abstract
본 발명은 항암제인 아클라시노마이신들과 그의 아글리콘을 생산하는 스트렙토마이세스속 균주 및 이를 배양하여 아클라시노마이신들과 그의 아글리콘을 생산하는 방법을 제공한다. 본 발명의 균주 스트렙토마이세스 라벤도폴리아(Streptomyces lavendofoliae) DKRs(KCTC 8539P)는 아클라시노마이신 A와 B만을 생산하는 친주인 I2/3A와 달리 A, B, Y 및 아글리콘을 생산하며 그 수율도 높다.
한편 미생물 배양 후 배양액을 염산이나 초산 완충용액으로 처리하면 아클라시노마이신 A가 Y로 또는 Y가 A로 전환되는 것이 관찰되는데, 이는 미생물 자체의 트정 효소의 작용에 의해 일어나는 것으로 생각되며, 이런 작용을 이용하여 아클라시노마이신 A 또는 Y를 선택적으로 생산할 수 있다.

Claims (7)
- 아클라시노마이신 A, B, Y 및 아글리콘의 생산기능을 갖는 스트렙토마이세스 라벤도폴리아(Streptolmyces lavendofoliae) DKRs(KCTC 8539P).
- 제1항에 있어서, 배양후 배양액을 초산 완충액으로 pH 4.4로 보정하면 아클라시노마이신 A를 Y로 전환시키고, 배양액을 염산으로 pH 4.6으로 보정하면 아클라시노마신 Y를 A로 전환시키는 능력을 가짐을 특징으로 하는 스트렙토마이세스 라벤도폴리아(Streptomyces lavendofoliae) DKRS(KCTC 8539P).
- 스트렙토마이세스 라벤도폴리아(Streptomyces lavendofoliae) DKRS(KCTC 8539P)를 배지에 배양하여 아클라시노마이신 A, B, Y 및 아글리콘을 균체내 및 배양액에 축적한 후 균체 및 배양액으로부터 아클라시노마이신 A, B, Y 및 아글리콘을 회수하여 정제함을 특징으로 하는 발효법에 아클라시노마이신 A, B, Y 및 아글리콘의 제조방법.
- 제3항에 있어서, 배양은 28-30℃, pH6.8-7.5범위의 호기적 조건에서 4-5일간 실시함을 특징으로 하는 제조방법.
- 제3항에 있어서, 톨루엔과 이소프로파놀의 30 : 1-35 : 1부피비의 혼합 용매를 이용하여 아클라시노마이신 A, B, Y 및 아글리콘을 회수 및 정제함을 특징으로 하는 제조방법.
- 스트렙토마이세스 라벤도폴리아(Streptomyces lavendofoliae) DKRS(KCTC 8539P)를 배지에 배양하고, 배양액을 초산 완충용액으로 pH 4.4로 보정한 후 균체와 배양액으로부터 아클라시노마이신 Y를 분리함을 특징으로 하는 아클라시노마이신 Y의 제조방법.
- 스트렙토마이세스 라벤도폴리아(Streptomyces lavendofoliae) DKRS(KCTC 8539P)를 배지에배양하고, 배양액을 염산으로 pH 4.6으로 보정한 후 균체와 배양액으로부터 아클라시노마이신 A를 분리함을 특징으로 하는 아클라시노마이신 A의 제조방법.※ 참고사항 : 최초출원 내용에 의하여 공개하는 것임.
