Abstract

PURPOSE:To achieve a highly sensitive and accurate diagnosis blocking nonpeculiar agglutination of a latex reagent by treating a serum containing an antibody after sensitization of the antibody to a latex particle. CONSTITUTION:An antibody is sensitized to a latex particle with a grain size of 0.01-1mu of a polystylene, stylene butadiene copolymer or the like in a weak alkali phosphoric acid buffer solution to obtain a sensitized latex removing unadsorbed antibody. The sensitized latex is incubated in a liquid in which the PBS is added to an anti-serum immunized by an antigen. The latex is centrifugally separated and redispersed in a PBS of about pH7 to obtain a latex reagent. This enables an accurate diagnosis without causing nonpeculiar agglutination tending to block reaction between an antigen and antibody in a serum being inspected. Thus, antigen on the surface of B hepatitis virus, for example, can be detected with a sensitivity about 1,000 times as large as that of the conventional reagent.
